When using different buffers in the gel (discontinuous gel electrophoresis), the gels are made up to one day prior to electrophoresis, so that the diffusion does not lead to a mixing of the buffers. By adding the catalyst TEMED and the radical initiator ammonium persulfate (APS) the polymerisation is started. The solution is then poured between the glass plates without creating bubbles. Depending on the amount of catalyst and radical starter and depending on the temperature, the polymerisation lasts between a quarter of an hour and several hours. === N-degrons === Proteolytic cleavage can lead to exposure of residues at the N-terminus of a protein. According to the N-end rule, different N-terminal amino acids (or N-degrons) are recognized to a different extent by their appropriate ubiquitin ligase (N-recognin), influencing the half-life of the protein. == Background == In 2012, Mali experienced a deep political and security crisis due to the insurgency in its northern regions. A military coup in Bamako and the rise of armed Tuareg groups, such as the National Movement for the Liberation of Azawad (MNLA) and jihadist elements, destabilised the region further. By 2013, the Malian government, supported by French forces under Operation Serval, as well as troops from Chad and other African nations, managed to recapture much of the north from jihadist control. However, the MNLA had taken advantage of the power vacuum left by retreating jihadists to regain control over key cities in northern Mali, particularly Kidal, a strategic town in the region of Azawad. In this context, tensions escalated between the MNLA and the Malian government over control of Kidal and the organisation of the upcoming presidential elections. While initially refusing to allow elections in Kidal, the MNLA eventually agreed to permit them in July 2013, provided that United Nations troops secured the voting process across Azawad. The Malian government, however, insisted that its own military forces must be present in Kidal to oversee security, which led to further friction between the two parties. By June 2013, tensions boiled over when the MNLA arrested 180 individuals in Kidal, accusing them of being Malian spies or infiltrators. Accusations of violence, pillaging, and ethnic targeting by the MNLA against black populations such as the Songhai, Peulh, and Bella people, were made.
==== Binding capacity ==== Proponents of both agarose and magnetic beads can argue whether the vast difference in the binding capacities of the two beads favors one particular type of bead. In a bead-to-bead comparison, agarose beads have significantly greater surface area and therefore a greater binding capacity than magnetic beads due to the large bead size and sponge-like structure. But the variable pore size of the agarose causes a potential upper size limit that may affect the binding of extremely large proteins or protein complexes to internal binding sites, and therefore magnetic beads may be better suited for immunoprecipitating large proteins or protein complexes than agarose beads, although there is a lack of independent comparative evidence that proves either case. Some argue that the significantly greater binding capacity of agarose beads may be a disadvantage because of the larger capacity of non-specific binding. Others may argue for the use of magnetic beads because of the greater quantity of antibody required to saturate the total binding capacity of agarose beads, which would obviously be an economical disadvantage of using agarose. While these arguments are correct outside the context of their practical use, these lines of reasoning ignore two key aspects of the principle of immunoprecipitation that demonstrates that the decision to use agarose or magnetic beads is not simply determined by binding capacity.
