Everything below concerns reversed-phase HPLC. We keep the language plain, cite what the science says, and separate well-supported claims from open questions.

Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.
Quality control relies on pharmacopoeial monographs where they exist, combined with in-house specifications for identity, purity, water content, and counter-ion composition. Reference standards allow calibration across laboratories, although certified materials for every analogue are not universally obtainable. Batch records, chromatograms, and mass spectra form the documentation trail. Regulatory classification varies by jurisdiction and intended use, and research-grade material differs from pharmaceutical-grade material in testing scope. Analytical uncertainty is often expressed as relative standard deviation across replicate injections.
Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.

Quantification and purity assessment rely on separation methods coupled to optical or mass detection. Reversed-phase high-performance liquid chromatography resolves the intact peptide from related impurities and is the standard assay technique. Size-exclusion chromatography measures aggregates, while ion-exchange chromatography separates charge variants produced by deamidation. Mass spectrometry confirms identity and detects mass shifts of a few daltons. In biological matrices, liquid chromatography with tandem mass spectrometry is often used because immunoassays can cross-react with endogenous GLP-1 or with circulating fragments.
As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
Stability studies focus on deamidation of asparagine and glutamine residues, oxidation of methionine, and aggregation into higher-order species. The fatty acid side chain adds susceptibility to oxidative change and can promote self-association at high concentration. Lyophilised material is comparatively robust when kept cold and dry, while aqueous solutions require refrigeration and protection from light. Forced degradation experiments under heat, acid, base, and peroxide conditions establish the specificity of each analytical method. Which degradation route dominates under real storage conditions depends on the formulation and stays formulation-specific.
Handling guidance for research quantities calls for single-use aliquots, an inert atmosphere where practical, and avoidance of repeated freeze-thaw cycles that accelerate aggregation. Certificates of analysis typically report purity by peak area, water content, counter-ion identity, and residual solvent levels. In the scientific literature the compound is usually described by its full amino acid sequence, its registry number, or its structural class rather than by any proprietary label. Reporting standards vary between journals, and reviewers increasingly request raw chromatograms alongside tabulated purity figures. Whether current purity thresholds are adequate for every experimental context is debated.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
==== Congress of South African Students ==== Accounts of the uprising typically accord a central role to black young adults – known in the anti-apartheid movement as "young lions" or "comrades" – who became the "shock troops" of the revolt. Their largest and most prominent organisational base was COSAS, another UDF affiliate that was founded in 1979, with a local branch in Vaal from March 1980. COSAS apparently played an important role in politicising the youth of Vaal, including by encouraging them to support the VCA, and many of the most militant participants in the uprising were formally or informally affiliated to COSAS.
The functions of AGPs in plant growth and development processes rely heavily on the incredible diversity of their glycan and protein backbone moieties. In particular, it is the AG polysaccharides that are most likely to be involved in development. Most of the biological roles of AGPs have been identified through T-DNA insertional mutants characterization of genes or enzymes involved in AGP glycosylation, primarily in Arabidopsis thaliana. The galt2-6 single mutants revealed some physiological phenotypes under normal growth conditions, including reduced root hair length and density, reduced seed set, reduced adherent seed coat mucilage, and premature senescence. However, galt2galt5 double mutants showed more severe and pleiotropic physiological phenotypes than the single mutants with respect to root hair length and density and seed coat mucilage. Similarly, hpgt1hpgt2hpgt3 triple mutants showed several pleiotropic phenotypes including longer lateral roots, increased root hair length and density, thicker roots, smaller rosette leaves, shorter petioles, shorter inflorescence stems, reduced fertility, and shorter siliques. In the case of GALT31A, it has been found to be essential for embryo development in Arabidopsis. A T-DNA insertion in the 9th exon of GALT31A resulted in embryo lethality of this mutant line. Meanwhile, knockout mutants for KNS4/UPEX1 have collapsed pollen grains and abnormal pollen exine structure and morphology. In addition, kns4 single mutants exhibited reduced fertility, confirming that KNS4/UPEX1 is critical for pollen viability and development.
=== Prevalence in rain, soil, water bodies, and air === In 2022, levels of at least four perfluoroalkyl acids (PFAAs) in rain water worldwide greatly exceeded the EPA's lifetime drinking water health advisories as well as comparable Danish, Dutch, and European Union safety standards, leading to the conclusion that "the global spread of these four PFAAs in the atmosphere has led to the planetary boundary for chemical pollution being exceeded". The most common PFAS found in the environment is Trifluoroacetic acid (TFA). Its presence is ubiquitous in the environment, especially in aquatic ecosystems, where it persists with increasing concentrations globally. It had been thought that PFAAs would eventually end up in the oceans, where they would be diluted over decades, but a field study published in 2021 by researchers at Stockholm University found that they are often transferred from water to air when waves reach land, are a significant source of air pollution, and eventually get into rain. The researchers concluded that pollution may impact large areas. Soil is also contaminated and the chemicals have been found in remote areas such as Antarctica. Soil contamination can result in higher levels of PFAS found in foods such as white rice, coffee, and animals reared on contaminated ground. In 2024, a worldwide study of 45,000 groundwater samples found that 31% of samples contained levels of PFAS that were harmful to human health; these samples were taken from areas not near any obvious source of contamination.
=== 23 April === Russia claimed to have captured the village of Ocheretyne, Donetsk Oblast, which was attributed to a rotational error by Ukrainian military commanders that left the sector mostly undefended and led to Russian forces advancing by five kilometers. The Ukrainian foreign ministry announced that it would suspend consular services to overseas Ukrainian men who were eligible for military service until 18 May, with the exception of those returning to Ukraine. The SBU arrested a resident of Kharkiv on suspicion of spying for Russia. UK Prime Minister Rishi Sunak announced a new military aid package for Ukraine that included 400 vehicles, 1,600 weapons and four million rounds of ammunition, along with £500 million in funding. The US Senate passed the bill authorising financial aid for Ukraine. The bill must now be signed into law by President Joe Biden.

=== Depression === Some evidence supports the use of buprenorphine for depression. Buprenorphine/samidorphan, a combination product of buprenorphine and samidorphan (a preferential μ-opioid receptor antagonist), appears useful for treatment-resistant depression. A buprenorphine implant (developmental code name SK-2110) is under development by Shenzhen ScienCare Pharmaceutical in China for the treatment of refractory major depressive disorder.
== Medical uses == In the US and the EU, adalimumab is indicated for the treatment of rheumatoid arthritis, juvenile idiopathic arthritis, psoriatic arthritis, ankylosing spondylitis, adult and pediatric Crohn's disease, ulcerative colitis, plaque psoriasis, hidradenitis suppurativa, and uveitis. In the EU it is also indicated for chronic aggressive progressive pulmonary and bone sarcoidosis.
==== Inhibition of DNA repair ==== Inhibition of DNA repair processes is considered one of main mechanism of inorganic arsenic genotoxicity. Nucleotide excision repair (NER) and base excision repair (BER) are the processes implicated in the repair of DNA base damage induced by ROS after arsenic exposure. In particular, the NER mechanism is the major pathway for repairing bulky distortions in DNA double helix, while the BER mechanism is mainly implicated in the repair of single strand breaks induced by ROS, but inorganic arsenic could also repress the BER mechanism. Exposure of isolated lymphocytes to arsenic causes decreased expression of the DNA repair protein ERCC1. Consistent with an inhibitory effect on DNA repair, lymphocytes from arsenic exposed individuals have higher levels of DNA damage. Arsenic can act as a co-carcinogen by inhibiting repair of DNA damage through its interaction with sensitive zinc finger DNA repair proteins.
During Operation Moduler, Cuban combat troops had remained well north of the Lomba River and declined to participate in the fighting, per Castro's instructions. In Luanda, President dos Santos summoned General Gusev and the senior Cuban general officer, Gustavo Fleitas Ramirez, for an urgent conference to discuss the worsening military situation and the failure of Operation Saluting October. Ramirez reminded dos Santos that Cuba had been opposed to the offensive from the beginning. Gusev lamented in his memoirs that "I informed [chief of the Soviet general staff] Akhromeyev about the result of the operation, but the most difficult task, in moral terms, was to inform the president of Angola, whom I had assured that the operation would succeed and that Savimbi would be crushed". On 25 November 1987, United Nations Security Council Resolution 602 was passed, condemning Operation Moduler as an illegal violation of Angolan sovereignty. The resolution expressed dismay at the continued presence of SADF troops in Angola and called for their unconditional withdrawal. South African foreign minister Pik Botha flatly dismissed the resolution out of hand, citing the unaddressed issue of Cuban linkage. He promised that the SADF would depart Angola once FAPLA's Cuban and Soviet advisers had likewise been withdrawn, or when their presence no longer threatened South African interests.
== Dynamic structures and modifications == As in the broader field of DNA nanotechnology, DNA origami may be made dynamic in nature through the use of a variety of methods. The three primary methods of creating a dynamic DNA origami machine are toehold mediated strand displacement, enzymatic reactions, and base stacking. While these methods are most commonly used, additional methods for creating dynamic DNA origami machines exist, such as designing a directional component and using brownian motion to drive rotational movement of structures or leveraging less commonly used DNA self-assembly phenomena like G-quadruplexes or i-motifs which can be pH sensitive.

== Criteria == While proteomic methods identified many novel proteins that potentially might be adhesome components they cannot be regarded as adhesome components until they are validated to fulfill the following criteria: 1. they localize to a cell adhesion structure such as focal adhesion or adherens junction. 2. they directly interact with one of the core adhesome components, such as integrin, cadherin or catenins AND/OR their knockdown has a clear effect on cell adhesion.
Half-Life: Counter-Strike spawned its own series which gradually became separate from the main Half-Life games, bar occasional references (such as an Easter egg referencing Portal present in Counter-Strike: Global Offensive).

=== The Nobel Prize in Chemistry 1944 === On 16 November 1945 the Royal Swedish Academy of Sciences announced that Hahn had been awarded the 1944 Nobel Prize in Chemistry "for his discovery of the fission of heavy atomic nuclei." Hahn was still at Farm Hall when the announcement was made; thus, his whereabouts were a secret, and it was impossible for the Nobel committee to send him a congratulatory telegram. Instead, he learned about his award on 18 November through the Daily Telegraph. His fellow interned scientists celebrated his award by giving speeches, making jokes, and composing songs. Hahn had been nominated for the chemistry and the physics Nobel prizes many times even before the discovery of nuclear fission. Several more followed for the discovery of fission. The Nobel prize nominations were vetted by committees of five, one for each award. Although Hahn and Meitner received nominations for physics, radioactivity and radioactive elements had traditionally been seen as the domain of chemistry, and so the Nobel Committee for Chemistry evaluated the nominations. The committee received reports from Theodor Svedberg and Arne Westgren. These chemists were impressed by Hahn's work, but felt that of Meitner and Frisch was not extraordinary, and did not understand why the physics community regarded their work as seminal. As for Strassmann, although his name was on the papers, there was a long-standing policy of conferring awards on the most senior scientist in a collaboration. The committee therefore recommended that Hahn alone be given the chemistry prize.

Reverse-phase high-performance liquid chromatography with ultraviolet detection is the usual choice, with results reported as area percent. Complementary methods such as size-exclusion chromatography and mass spectrometry are needed because a single separation cannot resolve every impurity class. Purity figures are therefore method dependent and should always be read alongside the technique used.
Oxidation mainly affects methionine residues and is promoted by dissolved oxygen, trace transition metals, and prolonged exposure to light. Buffer choice and the presence of antioxidants in a formulation can alter the rate appreciably. Because the products differ in mass by only a few units, mass spectrometry is often required to detect them.
Published data on long-term ambient stability are limited, so the question remains open rather than settled. Short excursions during transport are common in practice, and many suppliers use insulated packaging with cold packs. Where stability data are absent, cold-chain handling with temperature logging is the safer approach.
The lyophilized powder is dissolved in a suitable solvent, often sterile water or a buffered diluent, with gentle mixing rather than vigorous shaking. Foaming and shear should be avoided because they can promote aggregation. The resulting solution is then stored cold and protected from light.
