The result is a very strong coffee concentrate that is usually mixed with milk and sweetened. Many coffee retailers simply use hot-brewed coffee in their iced coffee drinks. Starbucks specifically uses the double-strength method in which the coffee is brewed hot with twice the amount of grounds. With this method, the melted ice does not dilute the strength and flavor of the coffee. Unlike the cold-brew process, this method does not eliminate the acidity inherent in hot-brewed coffee.
=== Climate change === Dyson agreed that technically humans and additional CO2 emissions contribute to warming. However, he felt that the benefits of additional CO2 outweighed any associated negative effects. He said that in many ways increased atmospheric carbon dioxide is beneficial, and that it is increasing biological growth, agricultural yields and forests. He believed that existing simulation models of climate change fail to account for some important factors, and that the results thus contain too great a margin of error to reliably predict trends. He argued that political efforts to reduce the causes of climate change distract from other global problems that should take priority, and viewed the acceptance of climate change as comparable to religion. In 2009, Dyson criticised James Hansen's climate-change activism. "The person who is really responsible for this overestimate of global warming is Jim Hansen. He consistently exaggerates all the dangers... Hansen has turned his science into ideology." Hansen responded that Dyson "doesn't know what he's talking about... If he's going to wander into something with major consequences for humanity and other life on the planet, then he should first do his homework- which he obviously has not done on global warming".
=== Technology in government === Newsom released his first book, Citizenville: How to Take the Town Square Digital and Reinvent Government, on February 7, 2013. The book discusses the Gov 2.0 movement taking place across the nation. After its release, Newsom began to work with the Center for Information Technology Research in the Interest of Society at the University of California, Berkeley, on the California Report Card (CRC). The CRC is a mobile-optimized platform that allows state residents to "grade" their state on six timely issues. The CRC exemplifies ideas presented in Citizenville, encouraging direct public involvement in government affairs via technology. In 2015, Newsom partnered with the Institute for Advanced Technology and Public Policy at California Polytechnic State University to launch Digital Democracy, an online tool that uses facial and voice recognition to enable users to navigate California legislative proceedings.
== Function == Cavin-2 is required for blood vessel formation (angiogenesis) in humans and zebrafish and required also for the endothelial cell proliferation, migration and invasion in humans. Cavin-2 plays an important role in endothelial cell maintenance by regulating eNOS activity. Cavin-2 controls the generation of nitric oxide (NO) in human endothelial cells by controlling the activity and stability of the protein endothelial nitric-oxide synthase (eNOS).

==== Cell proliferation ==== In order to have the mass differentiation of cells needed to form the specified cell tissues of different organisms, proliferation and growth of embryonic stem cells must take place. This process is mediated through canonical Wnt signaling, which increases nuclear and cytoplasmic β-catenin. Increased β-catenin can initiate transcriptional activation of proteins such as cyclin D1 and c-myc, which control the G1 to S phase transition in the cell cycle. Entry into the S phase causes DNA replication and ultimately mitosis, which are responsible for cell proliferation. This proliferation increase is directly paired with cell differentiation because as the stem cells proliferate, they also differentiate. This allows for overall growth and development of specific tissue systems during embryonic development. This is apparent in systems such as the circulatory system where Wnt3a leads to proliferation and expansion of hematopoietic stem cells needed for red blood cell formation. The biochemistry of cancer stem cells is subtly different from that of other tumor cells. These so-called Wnt-addicted cells hijack and depend on constant stimulation of the Wnt pathway to promote their uncontrolled growth, survival and migration. In cancer, Wnt signaling can become independent of regular stimuli, through mutations in downstream oncogenes and tumor suppressor genes that become permanently activated even though the normal receptor has not received a signal.
=== Winning streak === Next, Pudzian faced former bodybuilder Erko Jun on 9 November 2019 at KSW 51: Croatia. He won the bout via second round TKO. Pudzianowski was then scheduled to headline KSW 53 against Quentin Domingos on 21 March 2020 before the bout was scrapped due to Pudzianowski's injury. Pudzianowski was set to return after a fifteen month layoff against the undefeated Senegalese heavyweight Serigne Ousmane at KSW 59: Fight Code on 20 March 2021. However, on the day of the fight, Ousmane Dia suffered an acute appendicitis attack and had to be hospitalized. Stepping in on just a few hours notice was Serbia's Nikola Milanovic. Pudzianowski won the bout via TKO in the first round. Pudzianowski faced KSW 1 tournament winner Łukasz Jurkowski on 5 June 2021 at KSW 63: Crime of The Century. Pudzianowski won the fight via TKO in the 3rd round. The fight with Serigne Ousmane Dia was then rebooked and took place at KSW 64: Przybysz vs. Santos on 23 October 2021. Pudzianowski won the fight via knockout 18 seconds into round one. Pudzianowski faced former KSW Middleweight champion Michał Materla at KSW 70: Pudzianowski vs. Materla on 28 May 2022. He won the bout in the first round, knocking out Materla with an uppercut. He was awarded the Knockout of the Night bonus with the win. Among these, the recently described (2023) fungus Tremella parietinae is found exclusively on X. parietina, where it induces the formation of convex, orange to yellow galls within the hymenium of its apothecia. This fungus is known from several European countries—including Austria, Spain, and Sweden—and likely has a broader distribution wherever its host occurs. Another lichenicolous fungus, Tremella occultixanthoriae, parasitizes X. parietina but develops on the lower surface of the thallus and produces four-celled basidia. Both fungi belong to a broader complex of Tremella species that specialize in infecting Teloschistaceae lichens, often with strict host specificity. Some common saprotrophic fungi not typically associated with lichens have also been found colonizing X. parietina. These include Epicoccum nigrum, appearing as dark purple-brown hemispherical heaps on the surface of apothecial discs, and Periconia digitata, which forms distinctive stalked spherical heads resembling tiny pins on the apothecia and thallus. The environmental conditions that trigger such extensive fungal colonization remain unclear, although factors like prolonged snow cover, high rainfall, or other environmental stressors may initially weaken the lichen, making it susceptible to fungal invasion. Another lichenicolous fungus is Arthonia parietinaria, which was long misidentified as Arthonia molendoi or A. epiphyscia. This species forms mat black, dull ascomata that are arranged in large groups of up to 20–30 (sometimes 50) on the thallus surface of X.
== Efficacy and susceptibility == Cefodizime demonstrated superior clinical efficacy that what could be predicted from its in vitro experimental results. Certain immunomodulatory properties of the drug might account for its increased in vivo activity against specific bacterial species. These immunomodulatory properties stimulate some phagocyte and lymphocyte cellular functions when they have been impaired. It was shown in a 2004 study that cefodizime increased expression of toll-like receptor 4 (TLR-4) in neutrophils stimulated by Klebsiella pneumoniae infection which then affected the production of cytokines in the stimulated cells. A 1992 study conducted in vitro susceptibility studies for cefodizime and found that gram negative bacteria with consistent susceptibility include: Enterobacteriaceae including E. coli, K. pneumoniae, Morganella morganii, Proteus mirabilis, P. vulgaris, Shigella sonnei, Yersinia enterocolitica, Salmonella, Haemophilus influenzae, Moraxella catarrhalis, Neisseria gonorrhoeae, and Neisseria meningitidis. In vitro susceptible gram positive bacteria include: methicillin-sensitive Staphylococcus aureus, and Streptococcus agalactiae, S. pneumoniae and S. pyogenes. Cefodizime was shown to be ineffective in vitro against Enterobacter spp., Pseudomonas aeruginosa, and Xanthomonas maltophilia. Gram positive bacteria it was inactive against include Staphylococcus epidermidis and methicillin-resistant Streptococcus aureus.
Inspired by Michael Abrash, who left Microsoft to work on the game Quake at id, Newell and another employee, Mike Harrington, left Microsoft to found the video game company Valve on August 24, 1996. Newell opted to found Valve instead of retiring as he felt working with "other really smart, motivated, socially orientated people to create product that would affect millions of other people" would be more fun. Newell and Harrington funded development of the first Valve game, the first-person shooter Half-Life (1998), which was a critical and commercial success. Harrington sold his stake in Valve to Newell in 2000. Newell gave Valve no deadline and a "virtually unlimited" budget to develop Half-Life 2 (2004), promising to fund it himself if necessary. He and Valve came close to bankruptcy during a legal battle with Vivendi Games, which ended when an intern discovered an email revealing that Vivendi was destroying evidence.

Despite years of study, there are many various varieties and numbers of promoters and terminators for Escherichia coli, but also for the well-researched model organism Saccharomyces cerevisiae, as well as for other organisms of interest, these tools are quite scarce. === Electrostatic and hydrophobic interactions === Electrostatic interactions can form nanogels through the combination of anionic and cationic polymers in an aqueous solution. The size and surface charge of the resulting nanogels can be modulated by changing the molecular weight or the charge ratio of the two different polymers. Ionotropic gelation can also leverage electrostatic interactions between multivalent anions and cations to form nanogels. Hydrophobic interactions rely heavily on physical crosslinking to form nanogels. In this method, hydrophobic groups are added to hydrophilic polymers in an aqueous solution to induce their self-assembly into nanogels. When thiolated polymers (thiomers) are used for this preparation process, nanogels can be further stabilized by the formation of inter- and intrachain disulfide bonds due to oxidation. In the following the oppositely charged oligo- or polymers can even be removed.
