Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.

Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
Receptor binding triggers G protein signaling that raises intracellular cyclic AMP in pancreatic beta cells. Insulin release follows in a glucose-dependent manner, so secretion increases when blood glucose is elevated and diminishes when it is not. The same signaling suppresses glucagon release from alpha cells and slows gastric emptying, which blunts the post-meal glucose rise. In the brain, receptor activation in regions such as the arcuate nucleus is associated with reduced appetite and lower energy intake. How much each of these effects contributes to overall weight change is not fully settled.
Two structural features account for the prolonged half-life of semaglutide. A modified amino acid at position 8 resists cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GLP-1. A fatty diacid side chain binds serum albumin, which limits renal clearance and protects the peptide from enzymatic breakdown. These modifications yield a plasma half-life of approximately one week in humans, allowing once-weekly administration. The relationship between plasma concentration and clinical effect varies between individuals, and sources of that variability are still being characterized.
Semaglutide is a synthetic peptide analog of glucagon-like peptide-1 (GLP-1), a hormone released from intestinal L-cells after food intake. The compound belongs to the incretin mimetic class and acts at GLP-1 receptors distributed across pancreatic, gastrointestinal, cardiovascular, and central nervous system tissues. Compared with native GLP-1, the molecule carries structural changes that extend its activity from minutes to roughly one week. It is studied for glycemic control in type 2 diabetes and for weight management, and its effects on cardiovascular and other outcomes remain active research areas.
Receptor activation follows the canonical Gs pathway: binding increases intracellular cyclic AMP, which promotes protein kinase A activity. In pancreatic beta cells this amplifies glucose-dependent insulin release, so secretion rises when blood glucose is high and changes little when it is low. The same signalling suppresses glucagon release from alpha cells and slows gastric emptying. Receptors in the hypothalamus and brainstem are thought to contribute to reduced appetite and lower energy intake. Which of these effects dominates clinical outcomes remains an area of active study.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
The United States' efforts to assert influence over the region, particularly regarding the construction and control of the Panama Canal, contributed to the secession of the Department of Panama in 1903 and its political independence. The United States paid Colombia $25,000,000 in 1921, seven years after completion of the canal, as redress for President Theodore Roosevelt's role in the creation of Panama, and Colombia recognized Panama under the terms of the Thomson–Urrutia Treaty. Colombia and Peru went to war because of territory disputes far in the Amazon basin. The war ended with a peace deal brokered by the League of Nations. The League finally awarded the disputed area to Colombia in June 1934.
Like many other anemones, S. helianthus can serve as a “hub” for mutualistic networks of species and has been documented to live in symbiosis with other organisms. Symbiosis refers to the close association between organisms of two different species, whereas at least one organism benefits. These symbiont organisms vary substantially and can include algal endosymbionts, such as zooxanthellae, anemone crabs and anemone shrimp. In specific, Symbiodinium spp. is zooxanthellae that has been investigated to establish complex relationships with sun anemones, even to the extent of coordinating cell cycles with hosts. Another endosymbiont is the Clibanarius tricolor, or the blue-legged hermit crab, which has been recorded to live in symbiosis in the Caribbean and Indo-Pacific Regions. This crab uses the surface of the anemone as a “microhabitat” and is protected from the anemone's harmful toxins by removable coverings on the surface of their bodies. Another largely-studied endosymbiont is the anemone shrimp. Hayes et al. reported cohabitation of sun anemones by Periclimenes rathbunae, also called sun anemone shrimp, in a random manner that may be directly influenced by anemone size in the West Indies. In a further study, Periclimenes yucatanicus, or spotted cleaner shrimp, were additionally observed in symbiosis with S. helianthus in the Florida Bay.
karyogram A karyotype which depicts the entire set of chromosomes in a cell or organism by using photomicrographs of the actual chromosomes as they appear in vivo (usually during metaphase, in their most condensed forms), as opposed to the idealized illustrations of chromosomes used in idiograms. The photomicrographs are often still arranged in pairs and by size for easier identification of particular chromosomes, whereas in the actual nucleus there is seldom any apparent organization.
== Nixon's visit to Moscow == At the time of the Easter Offensive, Kissinger was deeply involved in planning for Nixon's visit to Moscow in May 1972. The offensive brought to the fore the differences between Nixon and Kissinger. Nixon threatened to cancel his summit with Leonid Brezhnev in Moscow if the Soviet Union did not force North Vietnam to end the Easter Offensive at once, saying: "Whatever else happens, we cannot lose this war. The summit isn't worth a damn if the price for it is losing in Vietnam". Nixon, in his instructions to Kissinger, stated that he viewed the relations with the Soviet Union through the prism of the Vietnam War, and if the Soviets were not prepared to help, Kissinger "should just pack up and come home". Kissinger, for his part, believed that Nixon was massively exaggerating Soviet influence in North Vietnam and no longer believed, if he ever did, in Nixon's "linkage" concept. Kissinger feared that Nixon was obsessed with Vietnam, and damaging relations with the Soviet Union over Vietnam would destabilize the international power balance by increasing American-Soviet tensions. On 20 April 1972, Kissinger arrived in Moscow without informing the U.S. ambassador Jacob D. Beam, and then went to have tea with Brezhnev in the Kremlin. Nixon, as usual when under stress, departed for a marathon drinking session with Rebozo at Camp David, and via Haig kept sending messages to Kissinger urging him to be tough with Brezhnev.

== Therapeutic implications == A bradykinin-potentiating factor (BPF) which increases both the duration and magnitude of the effects of bradykinin on vasodilation and the consequent fall in blood pressure, was discovered in Bothrops jararaca venom. On the basis of this finding, a non-protein analog of BPF which was effective orally was developed: the first angiotensin converting enzyme inhibitor captopril. It was approved by the FDA for the treatment of hypertension in 1981. Currently, bradykinin inhibitors (antagonists) are being developed as potential therapies for hereditary angioedema. Icatibant is one such inhibitor. Additional bradykinin inhibitors exist. It has long been known in animal studies that bromelain, a substance obtained from the stems and leaves of the pineapple plant, suppresses trauma-induced swelling caused by the release of bradykinin into the bloodstream and tissues. Other substances that act as bradykinin inhibitors include aloe and polyphenols, substances found in red wine and green tea.
=== Fermentation === Possibly the world's oldest fermented beverage, dating from 9,000 years ago, mead ("honey wine") is the alcoholic product made by adding yeast to honey-water must and fermenting it for weeks or months. The yeast Saccharomyces cerevisiae is commonly used in modern mead production. Mead varieties include drinks called metheglin (with spices or herbs), melomel (with fruit juices, such as grape, specifically called pyment), hippocras (with cinnamon), and sack mead (high concentration of honey), many of which have been developed as commercial products numbering in the hundreds in the United States. Honey is also used to make mead beer, called "braggot".
A study presented at the annual meeting of the American Geophysical Union in December 2006 asserted that a small-scale regional nuclear war could produce as many direct fatalities as all of World War II and disrupt the global climate for a decade or more. In a regional nuclear conflict scenario in which two opposing nations in the subtropics each used 50 Hiroshima-sized nuclear weapons (c. 15 kiloton each) on major population centers, the researchers predicted fatalities ranging from 2.6 million to 16.7 million per country. The authors of the study estimated that as much as five million tons of soot could be released, producing a cooling of several degrees over large areas of North America and Eurasia (including most of the grain-growing regions). The cooling would last for years and could be "catastrophic", according to the researchers. Either a limited or full-scale nuclear exchange could occur during an accidental nuclear war, in which the use of nuclear weapons is triggered unintentionally. Postulated triggers for this scenario have included malfunctioning early warning devices and/or targeting computers, deliberate malfeasance by rogue military commanders, consequences of an accidental straying of warplanes into enemy airspace, reactions to unannounced missile tests during tense diplomatic periods, reactions to military exercises, mistranslated or miscommunicated messages, and others. A number of these scenarios actually occurred during the Cold War, though none resulted in the use of nuclear weapons.

Garcia is a professor in the Departments of Molecular and Cellular Physiology and Structural Biology at the Stanford University School of Medicine, an Investigator of the Howard Hughes Medical Institute and a member of the National Academies of Science and Medicine. In addition to his role at Stanford, Garcia is a co-founder of several biotechnology companies, including Alexo Therapeutics, Surrozen, and 3T Biosciences.
=== Occurrence in fungi === Many fungi produce hydrogen cyanide from glycine. These include representatives of the genera funnel mushrooms (Clitocybe), dwindlers (Marasmius), stem porcini (Polyporus), and Ritterlinge (Tricholoma). The epurpurins are a group of yellow phenolic pigments, each bearing two nitrile groups, occurring in Emericella purpurea. Diatretin II occurs in Fleshy Fungus (Clitocybe diatreta) and in the purple reddish bolete. In the clove dwarf mushroom, the cyanohydrin of glyoxylic acid is present; it is formed from two glycine molecules and releases hydrocyanic acid upon tissue damage.
