Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.
Storage at minus 20 degrees Celsius or lower in a desiccated container preserves the peptide for extended periods, while working solutions are commonly held at two to eight degrees Celsius for short intervals. Light exposure and repeated freeze-thaw cycles accelerate degradation, so dividing material into single-use aliquots is generally recommended. Adsorption to glass and plastic surfaces can lower the measured concentration of dilute solutions, particularly below one milligram per millilitre. The degradation routes most often reported for GLP-1 analogues are deamidation, methionine oxidation, and backbone hydrolysis. Relative rates under specific conditions are frequently described only for individual formulations.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.

Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released by intestinal L cells after food intake. The natural hormone acts on pancreatic and central receptors but is degraded within minutes by dipeptidyl peptidase-4 and other peptidases. Semaglutide belongs to the class of long-acting GLP-1 receptor agonists, a group distinguished by structural changes that slow breakdown and extend circulation time. Its development followed earlier short-acting analogues and reflects a general strategy in peptide drug design: preserve receptor activity while blocking proteolytic clearance.
Three structural changes define the molecule. At position 8 an alpha-aminoisobutyric acid residue replaces alanine, which blocks dipeptidyl peptidase-4 cleavage. At position 34 arginine replaces lysine, and at position 26 a lysine carries a C18 fatty diacid attached through a short linker. The fatty chain binds serum albumin, and this albumin association reduces renal filtration and enzymatic attack. The unchanged backbone retains the receptor contacts that produce signalling. The free base has the formula C187H291N45O59 and a molecular weight near 4114 daltons.
=== Dehydration of amides and others === Nitriles can be prepared by the dehydration of primary amides. Common reagents for this include phosphorus pentoxide (P2O5) and thionyl chloride (SOCl2). In a related dehydration, secondary amides give nitriles by the von Braun amide degradation. In this case, one C-N bond is cleaved.
Yeast assimilable nitrogen or YAN is the combination of free amino nitrogen (FAN), ammonia (NH3) and ammonium (NH4+) that is available for a yeast, e.g. the wine yeast Saccharomyces cerevisiae, to use during fermentation. Outside of the fermentable sugars glucose and fructose, nitrogen is the most important nutrient needed to carry out a successful fermentation that doesn't end prior to the intended point of dryness or sees the development of off-odors and related wine faults. To this extent winemakers will often supplement the available YAN resources with nitrogen additives such as diammonium phosphate (DAP). However, the addition of excessive amounts of nitrogen can also create a hazard as other organisms besides beneficial wine yeast can utilize the nutrients. These include spoilage organisms such as Brettanomyces, Acetobacter and Lactic acid bacteria from the Lactobacillus and Pediococcus genera. This is why many wineries will measure the YAN after harvest and crushing using one of several methods available today including the nitrogen by o-phthaldialdehyde assay (NOPA) which requires the use of a spectrometer or the Formol titration method. Knowing the YAN in the must allows winemakers to calculate the right amount of additive needed to get through fermentation, leaving only "nutrient desert" for any spoilage organisms that come afterwards.

=== Antigen discrimination === A unique feature of T cells is their ability to discriminate between healthy and abnormal (e.g. infected or cancerous) cells in the body. Healthy cells typically express a large number of self derived pMHC on their cell surface and although the T cell antigen receptor can interact with at least a subset of these self pMHC, the T cell generally ignores these healthy cells. However, when these very same cells contain even minute quantities of pathogen derived pMHC, T cells are able to become activated and initiate immune responses. The ability of T cells to ignore healthy cells but respond when these same cells contain pathogen (or cancer) derived pMHC is known as antigen discrimination. The molecular mechanisms that underlie this process are controversial.

2.0 g beef extract 17.5 g casein hydrolysate 1.5 g starch 17.0 g agar 1 liter of distilled water. pH adjusted to neutral at 25 °C. Five percent sheep's blood and nicotinamide adenine dinucleotide may also be added when susceptibility testing is done on Streptococcus and Campylobacter species. It has a few properties that make it excellent for antibiotic use. First of all, it is a nonselective, nondifferential medium. This means that almost all organisms plated on it will grow. Additionally, it contains starch. Starch is known to absorb toxins released from bacteria, so that they cannot interfere with the antibiotics. Second, it is a loose agar. This allows for better diffusion of the antibiotics than most other plates. A better diffusion leads to a truer zone of inhibition. Mueller–Hinton agar was codeveloped by the microbiologist John Howard Mueller and the veterinary scientist Jane Hinton at Harvard University as a culture for gonococcus and meningococcus. They copublished the method in 1941.
Riots and protests resumed the following day, 17 December. The rioters broke into the district committee building and threw party documents, propaganda brochures, Ceaușescu's writings, and other symbols of Communist power out of windows. The military was sent in to control the riots, because the situation was beyond the capability of the Securitate and conventional police to handle. The presence of the army in the streets was an ominous sign; it meant that they had received their orders from the highest level of the command chain, presumably from Ceaușescu himself. The army failed to establish order, and chaos ensued, including gunfire, fights, casualties, and burned cars. Transportor Amfibiu Blindat (TAB) armoured personnel carriers and tanks were called in. After 20:00, from Piața Libertății (Liberty Square) to the Opera, there was wild shooting, including the area of Decebal bridge, Calea Lipovei (Lipovei Avenue) and Calea Girocului (Girocului Avenue). Tanks, trucks and TABs blocked the accesses into the city, while helicopters hovered overhead. After midnight, the protests calmed down. Colonel-General Ion Coman, local Party secretary Ilie Matei, and Colonel-General Ștefan Gușă (Chief of the Romanian General Staff) inspected the city. Some areas looked like the aftermath of a war: destruction, rubble and blood.
In December 2015, a norovirus outbreak at a Chipotle restaurant near Boston College sickened approximately 140 people, many of them students. The Boston Public Health Commission traced the outbreak to a sick employee who had been allowed to work. In response to the 2015 outbreaks, Chipotle closed all locations on February 8, 2016, for an all-staff food safety meeting and hired a new head of food safety who instituted enhanced protocols including more frequent handwashing, produce sanitation procedures, and Pascalization to pre-treat ingredients.

There are two closely related lep paralogues in Atlantic salmon (Salmo salar). A single lep gene has been documented for green-spotted pufferfish (Tetraodon nigroviridis), rainbow trout (Oncorhynchus mykiss), Arctic charr (Salvelinus alpinus), silver carp (Hypophthalmichthys molitrix), and grass carp (Ctenopharyngodon idellus). In other species there are reports of two closely related lep paralogues, including common carp (Cyprinus carpio) and Atlantic salmon. More distantly related lep genes have been found in medaka (Oryzias latipes) and zebrafish (Danio rerio). At least 2 leptin genes (lepa and lepb) exist in the crown-clade (Fig. 1). Early findings have shown that lepa and lepb share low interspecies aa identity, and are argued to have arisen through whole genome duplication, which occurred early in the teleost lineage. The duplicity of genes has been described for Atlantic salmon, Japanese medaka, common carp and zebrafish. Both lep paralogues cluster with lepa, and therefore suggest that at least one or more form (lepb) may exist in this species, since it is tetraploid. However, previous attempts using genomic synteny have only found the putative genomic duplicates in medaka and zebrafish paralogue. Currently it remains unclear, whether lepb exists in other teleosts due to the degenerative nature of this paralogue.
January 1916: creation of Secret Inter-Party Citizen's Committee (pl. Tajny Międzypartyjny Komitet Obywatelski) formed by members of the German Reichstag of Polish nationality. July 1918: a network of local Citizens' Committees is created on area of Prussian Partition. 11 October 1918: Polish organizations in the German Empire publicise common documents in which they declare the will to create independent Polish state and, in effect, revolution. 9 November 1918: beginning of the German Revolution, which also occurred in Greater Poland. Poles organized secret military structures in the Poznań garrison, Jarocin and Inowrocław. 10 November 1918: events of the so-called Republic of Ostrów. 11 November 1918 Armistice signed, ending the hostilities of World War I. The Citizens' Guard (Straż Obywatelska), renamed a few days later to the People's Guard (Straż Ludowa), comes out from the underground. The mayor of Poznań (Posen), Ernst Wilms, is removed from office. German military authorities give permission for functioning of the People's Guard to keep peace in the Province of Posen. 12 November 1918 The Central Citizen's Committee, later renamed to the Supreme People's Council (Naczelna Rada Ludowa, NRL), creates temporary "government-in-waiting" Commission (Komisariat): Stanisław Adamski, Wojciech Korfanty, and Adam Poszwiński. Jarogniew Drwęski becomes the Mayor of Poznań. 13 November 1918 Commission of the High People's Council calls citizens of German portion of Poland to keep calm in spite of the revolution.
=== Magnetic === In 2015 researchers announced a graphene-based magnetic sensor 100 times more sensitive than an equivalent device based on silicon (7,000 volts per amp-tesla). The sensor substrate was hexagonal boron nitride. The sensors were based on the Hall effect, in which a magnetic field induces a Lorentz force on moving electric charge carriers, leading to deflection and a measurable Hall voltage. In the worst case graphene roughly matched a best case silicon design. In the best case graphene required lower source current and power requirements.
=== Tuberculosis === Mycobacterium tuberculosis infection, or tuberculosis, has been shown to result in increased levels of active TGF-β within the lung. Due to the broad range of suppressive effects of TGF-β on immune cells, computer modeling has predicted that TGF-β blockade may improve immune responses and infection outcome. Research in animal models has further shown that TGF-β impairs immune responses and elimination of TGF-β signaling results in an enhanced T cell response and lower bacterial burdens. Thus, therapies which block TGF-β may have the potential to improve therapy for tuberculosis.

It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
The side chain enables reversible binding to albumin in the bloodstream, which protects the peptide from rapid renal clearance. This extends the interval between administrations compared with unmodified GLP-1.
No. It shares much of the native sequence but carries substitutions and an added side chain. These changes increase stability against enzymatic breakdown.
Filtering removes particulate matter that can block columns or scatter light. A 0.22 micrometre membrane is typical, and the filter material should be checked for peptide adsorption.
